Step 1: establish that there is no prescription to obtain
People who search where to get MOTS-c usually begin by looking for a clinical route, and the honest first step is to close that door properly rather than to work around it. Step 5: confirm the measured mass against 2,174.6 Da
Purity says how much of the sample is one substance. Identity says which substance. Only mass spectrometry answers the second question, and it answers it by comparison: measured mass against the mass calculated from the sequence.
MOTS-c is MRWQEMGYIFYPRKLR, 16 residues, which gives an average mass of about 2,174.6 Da and a monoisotopic mass of about 2,173.1 Da. Those figures differ by roughly 1.5 Da, so a certificate printing one theoretical value without naming the convention has left out something you need. A deconvoluted electrospray measurement should sit within a few daltons of the correct target. A shift near 16 Da has an obvious candidate in this sequence, since it carries two methionines and methionine oxidises readily, and that is a conversation to have with the supplier rather than an automatic rejection.
Steps 6 and 7: mass balance, endotoxin, and the code at checkout
A 10 mg vial holds 10 mg of powder, and powder includes residual water and counterion. Karl Fischer titration reports the water. The salt form reports the rest: material purified by reversed-phase HPLC is often isolated as a trifluoroacetate salt, and MOTS-c carries three arginines, a lysine, and a free N-terminal amine, so it can hold several TFA equivalents. At three equivalents roughly 14 percent of the labelled mass is counterion. Steps 8 and 9: match the label, then log and store
Step 8 is the only check that cannot be done in advance, and it is the one most people skip. Read the lot number printed on the vial and compare it against the lot number on the certificate you were sent. If they match, the analysis describes your material. If the vial carries no lot number, or a different one, everything read in steps 4 through 6 described something else.
Step 9 is record keeping and handling. Log the lot, the receipt date, the condition of the cake in the vial, and the certificate reference together, because a year later that record is the only thing linking the two. Lyophilised peptide is held frozen, protected from light and moisture, and allowed to reach room temperature before opening so condensation does not enter the vial. Where the sequence still fails
Completing all nine steps establishes what is in the vial to the limit that documents allow, and that limit is real. A certificate cannot show that the sample analysed represents the lot unless sampling is described, and it says nothing about manufacture, because analysis happens after synthesis. There is also no pharmacopoeial monograph for MOTS-c, so no specification exists for a certificate to be judged compliant against.
The larger gap is the compound itself. MOTS-c was characterised by Lee and colleagues in Cell Metabolism in 2015 in cell culture and mice, and most of what followed is rodent work. The only human clinical programme was CB4211, a MOTS-c analogue rather than MOTS-c, which completed a Phase 1a/1b study of 88 participants (NCT03998514) in April 2021 before its sponsor wound down.
